Isoniazid is a colorless crystal or white crystalline powder with a slightly bitter taste. It is chemically stable and highly water-soluble, suitable for various pharmaceutical and chemical applications.
Its hydrazine group has strong reducing properties, slowly converting dissolved oxygen to hydrogen peroxide in alkaline conditions. This reaction, catalyzed by gold nanoparticles, forms a luminol-based luminescent system with analytical potential.

The amino group on the hydrazine moiety also enables versatile reactivity: it forms Schiff bases with active carbonyl compounds, and reacts with carboxylic acid derivatives or sulfonyl chlorides to produce acylated or sulfonyl isoniazid derivatives.

Isoniazid is a first-line anti-tuberculosis drug with high efficacy, low toxicity and cost-effectiveness. It also serves as an electroplating additive and pharmaceutical intermediate.

In chemical synthesis, it condenses with formaldehyde sodium bisulfite to form isoniazid methanesulfonic acid (CAS#13447-95-5), and reacts with formaldehyde to produce di-isoniazid. This straightforward process is typically carried out by adding formaldehyde to aqueous isoniazid solution at ~65°C for 30 minutes.
Condensation with vanillic acid in aqueous solution at ~95°C yields isoniazid hydrazone. All these derivatives have anti-tuberculosis activity: isoniazid itself is the most potent, while isoniazid hydrazone shows lower activity against Mycobacterium tuberculosis but reduced neurotoxicity and hepatotoxicity.
Isoniazid remains one of the most critical anti-tuberculosis agents for research and clinical use, with high selectivity and strong activity against tubercle bacilli. In vitro, concentrations as low as 0.025–0.05 mg/L inhibit bacterial growth, and higher concentrations are bactericidal against actively multiplying bacteria.
